This file contains the data of Figure 6.

Figure 6 shows the flocculation coefficient calculated from optical density measurements of sedimenting cells suspended in 50 mM sodium acetate buffer.
The flocculation assays were performed for the strains CEN.PK113-7D, IMK1061, IMX2897, IMX2912, IMX2913, IMX2914 and IMX2915.
All strains were grown as biological duplicates for both dark conditions.
IMK1061 was only tested under dark conditions. CEN.PK113-7D and IMX2897 were tested under light and dark conditions. For these three strains, all tests were done using biological duplicates.
For the remaining strains, the tests in dark were done using biological duplicates, while the tests in light were performed multiples times over several experiments, resulting in 14 biological replicates per strain.
Sedimentation tests were performed in technical triplicates. Ooptical density measurements were each performed in technical duplicates.

The data for each strain is found in a seperate tab.
In each tab, the data pertaining to the graph is seen at the top, with the values in light orange representing the data points used to generate the graph, while the values in blue represent the means and standard deviations depicted in the graph.

The raw data and calculations is marked in 5 steps.

1. Raw OD660 measurements from under the meniscus of sedimenting cells in glass tubes after 5 minutes of vertical incubation. each sedimentation experiment was performed in triplicate per samples taken during exponential growth from triplicate cultures of each strain under light and dark conditions.
Each sample is measured in technical duplicates. These values are used to calculate Value B for the flocculation coefficient formula.

2. The averages are calculated for the technical duplicates and a correction was done to account for the addition of 300 ul CaCl2 to the samples.

3. The optical density was measured for the deflocculated cells and multiplied with the dilution factor to obtain the value A used for calculating the flocculation coefficient.

4. The actual optical density for the values at step 2 is calculated by multiplying with the dilution factor.

5. The flocculation coefficent is calculated using the values from step 3 and 4, and the formula: F = 100% x (A-B)/A
